摘    要:目的]为改善养殖的微生态环境提供参考。方法]以无纺布为载体材料,采用PCR-DGGE技术比较了生物膜和水体微生物多样性的差异。结果]电泳分析表明,总共检测到46条不同的条带。第4天,生物基与水体样品的条带数差异不明显。第8天时,二者出现了不同条带。在10~18天,生物膜多样性指数一直低于水体,直到第18~20天才开始高于水体。第12天时生物膜上优势菌种普遍存在,但是部分开始出现衰减和增强的趋势。Band1、5、6分别是生物膜上第8、12、16天出现的特征性条带,除假单胞菌属外,其他结果均为未得到分离培养或者未经过命名的菌类。水样中的特征条带Band3、4同爱德华菌、弧菌属的相似性分别为94%、87%。结论]生物基载体可以改善池塘生物多样性。

Abstract:Objective] The study was to provide a base to improve the microecology environment of aquaculture. Method] With non-woven fabric as carrier material, the difference of microbial diversity of biomembrane and water body and biology base in culture pond was compared by PCR-DGGE technique. Result] Sum of 46 bands were detected by PCR-DGGE. On 4th day, the bands of biology base samples were not different from that of water samples. At 8th day, different bands were emerged among them. On 10th~18th day, the diversity index of biomembrane was lower than that of water and higher than that of water till 18~20 day. On 12th day, the dominant strains in biomembrane existed commonly, but part had a thread of decreasing or increasing. Band 1, 5, 6 on biomembrane was the characteristic bands on 8, 12, 16 day resp. Beside Pseudomonas lubricans, others were all fungoid without cultured or named. The characteristic band 3, 4 in water samples had 94% and 87% similarity with Edwardsiella ictaluri and Vibrio resp.. Conclusion] The carrier of biology base could improve the biology diversity in culture pond.
