白蜡种子发芽能力检测方法

摘    要:采用离体胚法和四唑染色法分别测定了白蜡(Fraxinus chinensis Roxb.)种子的生活力,并采用5种不同的处理方法测定了该种子的发芽率。结果表明:四唑染色法测定种子的生活力为66.0%,离体胚法测定的生活力为70.5%。发芽率测定中,清水浸种21 d或用200 mg.L-1赤霉素浸种60 h后白蜡种子的发芽率较高,分别为60.5%和59.5%。采用相同质量浓度的赤霉素分别浸种36、48 h后,发芽率分别为49.25%和53.5%,远低于前2个处理的发芽率结果。同批种子采用浸种1 d、再去除果翅的方法测得的发芽率仅为22%。从种子发芽能力各测定法历时天数来看,四唑染色法所需时间最短,约为8 d;清水浸种需要时间最长,为34 d;而离体胚法和赤霉素浸种60 h的处理分别需要15.0、17.5 d。可见,某些单位采用清水浸种1 d即进行发芽试验的方法不能准确测定种子的发芽率,赤霉素处理是白蜡种子发芽率测定的较好方法。

Abstract:Tetrazolium staining test and embryo culture method were applied to determine the viability of Fraxinus chinensis seeds.The germination percentage of the seeds was determined by five different methods.Results showed that the viability of F.chinensis seeds was 70.5% by the excised embryo test and 66.0% by the tetrazolium staining test.Higher germination percentages were obtained when the seeds were soaked in water for 21 d or in 200 mg·L-1 GA3 for 60 h,which were 60.5% and 59.5%,respectively.The germination percentages were lower when the seeds were soaked in 200 mg·L-1 GA3 for 36 h or 48 h,which were 49.25% and 53.55%,respectively.When the seeds from same batch were soaked in water for one day and then winged perianths were removed,the germination percentage was only 22%.The germination time was 8.0 d for the tetrazolium staining test,34.0 d for the seeds soaked in water for 21 d,15.0 d for the excised embryo test,and 17.5 d for the seeds soaked in GA3 for 60 h.The germination test after soaking seeds in water for one day could not accurately determine the germination percentage of seeds,while soaking the seeds in GA3 is a better method for the determination of germination percentage of F.chinensis seeds.
