摘    要:采用已构建的能表达致断奶仔猪腹泻(PWD)和仔猪水肿病(ED)大肠杆菌保护性抗原FedA-Stx 2e B-FaeG融合蛋白的重组菌BL21(pFSFaeG)和重组菌X4550(pFSFaeG),经IPTG诱导后制成抗原,经口服及灭活后制成油乳剂苗免疫小鼠后,检测IgG抗体水平的变化。结果表明:重组菌经灭活后免疫小鼠能够诱导产生针对上述3种保护性抗原的特异性抗体IgG。免疫效力试验结果显示,两种重组菌的最佳免疫途径均为灭活后经腹腔免疫。

Abstract:The recombinant strains BL21(pFSFaeG) and X4550(pFSFaeG),which expressed protective antigens of porcine pathogenic Escherichia coli,and activity of toxin was lost completely were constructed.The 5-week-old mice were immunized with live recombinant bacteria orally and with inactivated vaccine by the peritoneal route.Sera and feces were simultaneously collected from the vaccined mice on the 7,14 d post both the first and second immunization.The result indicated that serum IgG specific to F4,F18 and Stx 2e antigens could be stimulated in mice immunized with inactivated vaccine,but could not in all oral groups.The protection efficacy of mice immunized with inactivated recombinant strain against challenging of strain 107/86,C83907 and Stx 2e was much better than mice immunized with live recombinant bacteria orally.
