长春花内生真菌的分离及鉴定

摘    要:目的]为研究内生真菌与长春花悬浮细胞互作奠定基础。方法]从长春花茎杆韧皮部分离出内生真菌,对分离出的菌株进行鉴定,用PDA培养基、PDA培养基+长春花悬浮细胞的水提物对内生真菌进行液体培养,薄板层析检测内生真菌液体培养物的提取物及发酵液。结果]从常规PDA培养基中分离得到11株菌株,而从其他PDA培养基中没有分离到内生真菌。分离出的11株菌株的大型分生孢子呈纺锤形或镰刀形,其代谢产物通过薄层层析,没有检测到长春花生物碱。表明分离出的内生真菌本身不能直接合成长春花生物碱,而可能对生物碱的合成起促进作用。结论]从长春花中分离得到的内生真菌属镰刀菌属。

Isolation and Identification on Endophytic Fungus from Catharanthus roseus
Abstract:Objective] The study aimed to lay foundation for the research on interaction of endophytic fungus with suspension cell of Catharanthus roseus.Method] The endophytic fungus were isolated from phloem of C.roseus stem,the isolated endophytic fungus strains were identified and cultured in liquid with PDA medium and aqueous extracts from PDA medium + suspension cell of C.roseus.The extracts and the fermentation broth from liquid cultural substance of endophytic fungus were detected with the method of thin-layer chromatography.Result] 11 strains were isolated from conventional PDA medium,but there were no endophytic fungus being isolated from other PDA mediums.The macroconidium of the 11 isolated strains showed as spindle shape or sickle shape,in the metabolites of the strains,there was no C.roseus alkaloid being detected out with the method of thin layer chromatography,which indicated that the isolated endophytic fungus could not directly synthetize C.roseus alkaloid,but they possible had promotion on the synthesis of C.roseus alkaloid.Conclusion] The endophytic fungus isolated from C.roseus belongs to fusarium.
