RP-HPLC法测定陆英叶中绿原酸含量的研究

摘    要:建立反相高效液相色谱法测定了陆英叶中绿原酸的含量,同时采用光电二极管阵列检测器检测绿原酸色谱峰的纯度。色谱柱为Nova-PakC18(3.9 mm×150mm,4μm),流动相为水-甲醇-乙酸(881∶21∶,V/V),流速1.0ml/min,检测波长326nm,柱温25℃。绿原酸在0.832~4.16μg范围内线性关系良好(r=0.99992),检出限为0.4mg/L(3σ),平均回收率为97.1%。该法简便、准确、重现性好、线性范围宽,适于对含有绿原酸的药材进行质量评价。

Abstract:A determination method of chlorogenic acid in Sambucus chinensis Lindl.leaf by reversed-phase high performance liquid chromtograph was established and at the time the purity of chlorogenic acid peak was detected by means of Photodiode Array Detector(PAD).The chromatographic column,Nova-Pak C18(3.9 mm×150 mm,4 μm),water-methanol-acetic acid(88∶12∶1,V/V)mobile phase with 1.0 ml/min flow rate,the detected wavelength(326 nm) and the column temperature(25 ℃) were adopted.The calibration curve of chlorogenic acid was linear under the content of 0.832 ~4.16 μg,the correlation coefficient was over 0.999 92 and the detected limit was 0.4 mg/L(3σ).The average recovery of chlorogenic acid was 97.1 %.The method can be adapted to evaluate the quality of medicine that contains chlorogenic acid.
