摘    要:采用mRNA差异显示和SSR(微卫星)技术,比较了小麦品种丰抗8号及其幼胚培养突变系第R8代株系的基因编码区和非编码区的核苷酸序列,证明突变系的DNA序列存在大量的突变,突变类型为:嘌呤与嘧啶相互转换、核苷酸片段缺失、增加核苷酸片段.DNA序列的突变揭示了突变系农艺性状和生理生化方面变异的分子基础.

Abstract:To explore the molecular basis of somaclonal variation in wheat, we co mpared DNA coding region and un-coding region between wheat cultivar Fengkang 8 and its mutant line by mRNA difference display and SSR (Microsatellites). The mu tant line was selected from the R8 mutants generated by culturing immature embry os. The sequences of the cloned cDNA fragments and the SSR patterns of Fengkang 8 and the mutant line revealed that mutations had occurred in both coding and un -coding regions of the mutant's genome. The mutations consist of conversions bet ween pyridine and purine as well as deletion and/or addition of DNA fragments, w hich may have caused the agronomical variations of the mutant line.
