中国10个地方蛋鸭品种遗传多样性及动态保种的研究

摘    要:通过资源调查,查明了我国11个地方蛋鸭品种的保存现状,发现文登鸭已濒临灭绝,其他10个蛋鸭品种均处于正常保种状态。利用筛选出的28个多态性较好的微卫星标记,对我国10个地方蛋鸭品种的遗传多样性进行检测。根据等位基因频率计算各群体的平均基因杂合度、多态信息含量和群体间的Ds遗传距离。结果表明:28个微卫星位点中有26个微卫星位点在10个鸭群体中的多态信息含量均为高度多态,可作为有效的遗传标记用于各品种的遗传多样性和系统发生关系的分析。10个鸭品种中,平均基因杂合度为0.514~0.617,各品种的杂合度均较高,最高的为三穗鸭,最低的是金定鸭;用UPGMA法对Ds遗传距离进行聚类分析,结果10个蛋鸭品种被聚为3类,福建省的山麻鸭、莆田黑鸭、连城白鸭聚为一类;湖北省的荆江麻鸭、恩施麻鸭和贵州省的三穗鸭聚为一类;山东省的微山麻鸭、湖南省的攸县麻鸭和浙江省的绍兴鸭聚为一类。根据调查结果及遗传多样性检测数据,分析了10个地方蛋鸭品种的保存状态,并提出了保护建议。

Abstract:A survey of conservation actuality of eleven indigenous egg-duck breeds was carried out in this study.The results showed that Wendeng duck had been close to extinction.The rest ten egg-duck breeds were in the condition of normal conservation.Twenty eight microsatellite markers with medium or high polymorphisms were selected to detect the genetic diversity of ten egg-duck breeds.According to the allele frequencies of 28 microsatellite sites,mean heterozygosity,polymorphism information content and standard genetic distances(Ds) were calculated for each breed.The results showed that 26 of 28 microsatellite sites were high polymorphic,so the 26 microsatellite markers were effective markers for analysis of genetic relationship among duck breeds.The mean heterozygosity of 10 duck breeds was all over 0.5.The highest was in the Jingjiangma(0.617),and the lowest was in the Mianyangma.According to the results of investigation and the detected data of genetic diversity,the situation of conservation was analyzed and the advice of conservation about the ten indigenous eggduck breeds was given.
