摘    要:采用正交试验设计,研究了4种植物激素(6-BA、NAA、GA3、2,4-D)的4个水平对3个马铃薯品种(陇薯3号、陇薯6号、LK99)茎段愈伤组织生长和分化的影响,筛选出了适合不同基因型的愈伤诱导和芽分化培养基.试验结果表明,诱导愈伤的最佳激素配比,陇薯3号为MS+6-BA 2.5 mg/L+NAA 0.5 mg/L+GA35 mg/L+2,4-D 1 mg/L,陇薯6号为MS+6-BA 2.5 mg/L+NAA 0.5 mg/L+GA32.5 mg/L+2,4-D 0.5 mg/L,LK99为MS+6-BA 2.5mg/L+NAA 1 mg/L;诱导芽分化的最佳激素配比,陇薯3号、陇薯6号为MS+6-BA 3.5 mg/L+NAA 1 mg/L+GA310 mg/L,LK99为MS+6-BA 1.5 mg/L+NAA 0.5 mg/L+GA310 mg/L.

Abstract:The L_(16)(4~(5)) orthogonal design was adapted to select the optimum plant hormone combination for regeneration of potato stem.Four plant hormones(6-BA,NAA,GA_(3),2,4-D)were applied at four different levels to study their influence on the growth and differentiation of potato(Longshu No.3,Longshu No.6,LK99)stem calli.The results indicated that the optimum medium for callus induction from stem was MS 6-BA 2.5 mg/L NAA 0.5 mg/L GA_(3 )5 mg/L 2,4-D 1 mg/L for "Longshu No.3",MS 6-BA 2.5 mg/L NAA 0.5 mg/L GA_(3)2.5 mg/L 2,4-D 0.5 mg/L for "Longshu No.6" and MS 6-BA 2.5 mg/L NAA 1 mg/L for "LK99".The optimum media for inducing bud differentiation from stem calli were found to be MS 6-BA 3.5 mg/L NAA 1 mg/L GA_(3)10 mg/L for "Longshu No.3" and "Longshu No.6" and MS 6-BA 1.5 mg/L NAA 0.5 mg/L GA_(3)10 mg/L for "LK99".
