应用XANE技术研究瘤胃液中锌的相对浓度和状态

摘    要:应用XANES(X射线吸收近边结构)技术,研究了体外培养瘤胃上清液中锌的相对浓度和状态。以4头安装永久性瘤胃瘘管的杂交绵羊为供体动物,早饲后2h采集瘤胃液,混合后用于体外培养。以植酸、碳酸钙、微晶纤维素和苜蓿,配制成植酸高钙、植酸低钙、低纤维素和高纤维素4种培养底物,在4种底物中按200mg/kg等量锌分别添加ZnMet、ZnLys、ZnSO4和ZnO,共16个处理。体外培养24h,3500rpm离心分离上清液,65℃烘干24h,应用XANES(X射线吸收近边结构)技术测定其中锌浓度和状态。结果表明:在植酸高钙或不同水平纤维素组中,上清液中锌浓度有机锌明显优于无机锌,但在植酸低钙组中,有机锌与无机锌差别不大。归一化后,植酸钙组锌的谱线基本相同,吸收边位置也与文献中的9 659eV 相同,说明经过24h培养的上清液中不同锌源锌的化学状态相同;但纤维素组锌的谱线不同,说明纤维素改变了上清液中锌的状态。XANES可作为矿物元素营养研究的新技术。

Abstract:Mixed rumen fluid were collected from 4 sheep with permanent rumen fistula after being fed for 2h,  Zn relative concentration and state in supernatant fluid of the rumen fluid was investigated through in vitro incubation. Four kinds of culture substances, which had characteristic of phytic acid-higher calcium, phytic acid-lower calcium, higher cellulose and lower cellulose, was added in 200mg/kg Zn of ZnMet, ZnLys (organic zinc), ZnSO4 and ZnO (inorganic zinc). Culture substances were centrifuged at 3500r/min after 24h incubation, and the supernatant fluid was dried for 24h and then zinc in it was determined by using XANES (X-ray absorption near edge structure). The result is that organic zinc obviously prevailed inorganic zinc in the treatment of phytic acid with calcium-higher and different level of cellulose in increasing zinc concentration, but no significant difference was found in the treatment of phytic acid with calcium-lower. The normalized spectrums of phytic acid with calcium groups were basically similar for 4 kinds of zinc source, indicating that after being cultured for 24h in vitro, zinc chemical states of all treatments in supernatant fluid were similar with each other, and absorption edge positions were also 9 659eV as described in the literatures. But the chemical states of zinc were changed in treatments of different cellulose levels, suggesting that zinc state in the supernatant fluid could be changed after cellulose was added. It is concluded that XANES could be a new technique for study animal mineral element nutrition.
