苹果煤污病与蝇粪病病原菌分离技术

摘    要: 苹果煤污病和蝇粪病的病原菌生长在果实的表面,由于分离过程中易于受到其它真菌和细菌的污染使得按常规方法分离相当困难。本研究使用直接挑取菌体分离技术进行了尝试,分离成功率高。为了保证每个样品至少长出2~3个纯菌落,每次分离应该有15个重复。通过菌落的生长速度和菌落形态的一致性可以判断长出的菌落是否是目标菌。

Abstract:The contaminations make it difficult to isolate sooty blotch and flyspeck fungi from apple surface. In this research we used the technique to pick up the thallus directly from apple surface under a stereomicroscope. For each sample, 10-15 slant repeats were needed to insure at least two to three isolations could grow up. Compared with common methods repor- ted previously, this technique is of high efficiency and less contamination rate.
