黑斑病菌Diplocarpon rosae侵染对现代月季抗感不同杂交后代叶片超微结构的影响

摘    要:以现代月季(Rosa hybrida)抗感不同的杂交后代作为研究对象,测定黑斑病菌(Diplocarpon rosae)侵染后对叶片表皮细胞结构的影响。经扫描电镜观察,在病菌未侵染时叶片表皮细胞均正常,表皮有气孔分布,气孔椭圆形且饱满。但接种病菌6d后,抗病后代中的感病叶片表皮均有大量菌丝分布,有少数菌丝进入气孔内部;感病后代中的感病叶片表皮肿胀、气孔下陷、孢子附着在叶片表面。通过透射电镜观察,未接种月季黑斑病菌时叶片表皮细胞正常,接种病原菌之后抗病后代叶肉细胞内含较大、较多的嗜锇颗粒,叶绿体由椭圆形变为近圆球形,类囊体形态均一性降低,排列松散,定向发生紊乱;感病后代细胞内叶绿体整体瓦解,只保留了液泡和细胞核,且出现较多的空泡。随着病菌的进一步侵染,细胞质完全降解,导致细胞死亡。

Abstract:Two Rose hybrida offsprings were examined differing in black spot tolerance at the subcellular level. The subcellular structure had no difference between 2 materials under non-stressed conditions. But chloroplasts and mitochondria of leaf cells of both Rose hybrida offsprings showed ultrastructural changes after 6 days of black spot inoculation. Observing with scanning electron microscope, large number of mycelium scattered on the leaf epidermis of resistant offspring, a few hyphae got into the stomata; While the susceptible one’s epidermis swelled and stomata subsidented, spore were floating on the surface. Observing with transmission electron microscope, cells of resistant offspring contained much osmiophilic granules, the shape of chloroplasts changed from oval to round. Thylakoid arranged loosely, directly disordered and its morphology homogeneity was reduced. Chloroplast in cell of susceptible one collapsed, only the vacuole and nucleus were kept, and more vacuoles were appeared. With further infection of pathogenic bacteria, cytoplasmic degraded completely until cells died.
