| 摘 要： | 采用优球蛋白法和Sepharose-4B柱层析纯化南方鮎血清免疫球蛋白,SDS-PAGE结果表明,重链相对分子质量约为7.4×104,轻链相对分子质量约为2.6×104;用纯化蛋白免疫Balb/c小鼠制备多克隆抗体,ELISA测定多克隆抗体效价高达1∶216;Western-blotting结果显示多克隆抗体与免疫球蛋白重链和轻链都有反应,与不同科的沟鮎血清和不同目的草鱼血清无交叉反应.利用优球蛋白法和Sepharose-4B柱层析首次纯化南方鮎血清免疫球蛋白,与Sepharose-4B柱层析相比优球蛋白法更简单、快速;制备的多克隆抗体具有特异性,可做为以后进一步研究南方鮎的免疫系统和免疫防治的工具.

| Abstract: | Serum immunoglobulin from Silurus meridionalis Chen was purified by euglobulin precipitation and sepharose-4B gel column chromatography;SDS-PAGE was performed to examine the purity of the protein;polyclonal antibodies were prepared by repeatedly immunized Balb/c mouse with the euglobulin precipitation-purified protein;indirect enzyme-linked immunosorbent assay(ELISA) was used to detect the titer of the polyclonal antibodies;and Western-blotting was performed to assess the specificity of the mouse antiserum.... |
