紫斑牡丹幼胚离体培养试验

摘    要:以紫斑牡丹离体幼胚为试材,研究了胚龄分别为盛花后30 d、45 d6、0 d7、5 d的幼胚萌发率,选取萌发率高的盛花后75 d胚龄的种子于4℃下低温预处理,观察低温预处理对幼胚萌发的影响,并筛选较为适合的紫斑牡丹幼胚初代培养和增殖培养的培养基成分。结果表明:胚龄为盛花后75 d的种胚易萌发,萌发率为58.3%,经4℃低温预处理后萌发率可达到68.3%;诱导幼胚萌发与成苗的适宜培养基为MS(微量元素3/2)+1.0 mg.L-16-BA+0.2 mg.L-1IAA+20 g.L-1蔗糖+10 g.L-1葡萄糖+300 mg.L-1CH+0.1 g.L-1琼脂,萌发率和成苗率分别达到100%、98.3%;培养基MS+1.0 mg.L-16-BA+0.01 mg.L-1NAA+30 g.L-1蔗糖+0.1 g.L-1琼脂的增殖培养效果较好,增殖倍数可达5.7。

Abstract:Immature embryos of Paeonia rockii were used to study germinating rate of different embryonic age (30, 45, 60 and 75 days after florescence), observed the effect of germination on immature embryos which seeds were treated with pre-chilling (4 ℃), and screened the optimum medium of primary culture and multiplication of subculture. Results showed that germinating rate of immature embryos (75 days after florescence) was as high as 58.3 %, and it had 68.3 % for prechilling seeds in 4 ℃. MS (trace elements3/2) + 1.0 mg·L~(-1) 6-BA +0.2 mg·L~(-1) IAA +20 mg·L~(-1) sucrose+10 mg·L~(-1) glucose+300 mg·L~(-1)CH as the effective medium that germinating rate was 100 % and plantlet forming rate reached 98.3 %; the most optimum proliferate medium was MS+1.0 mg·L~(-1) 6-BA+0.01 mg·L~(-1) NAA+30 g·L~(-1) sucrose, the proliferation rate reached 5.7.
