一株犬种布鲁氏菌的分离与鉴定

摘    要:对从一只流产贵宾犬全血中分离到的一株布鲁氏菌进行了形态结构、培养特性、生化特性研究以及凝集试验鉴定和多种属聚合酶链式反应(AMOS-PCR)鉴定.结果表明,分离菌为革兰氏阴性菌,在胰际琼脂培养基生长并呈典型的粗糙型布鲁氏菌菌落,菌落能被结晶紫溶液染成紫色.分离菌在琉瑾和复红培养基上生长,不产生H2S,与粗糙型布鲁氏菌阳性血清凝集,与光滑型布鲁氏菌阳性血清无凝集.用AMOS-PCR方法对分离菌进行鉴定,该分离菌具有犬种布鲁氏菌特有条带.鉴定结果表明分离菌符合犬种布鲁氏菌特性,将其命名为B.canis GB1.

Isolation and identification for a strain of brucella canis from a poodle

Abstract:In this study, a Brucella isolated from whole blood of a poodle aborted, which was further identified by morphology, cultural characteristics, biochemical characteristics, agglutination test and AMOS-PCR. The isolate was gram-negative bacteria, which could grow on the Brucalla agar. It was a characteristic rough colony, and can be dyed purple by crystal violet solution. The isolated bacteria did not produce H2S, while it could grow on the culture medium of thionine and fuchsin. The bacterial could agglutinate the serum against rough-type Brucella, while it could not agglutinate the serum against smooth-type Brucella. AMOS-PCR was used to identify its species, and confirmed it belongs to the member of B.canis. The isolate was named B.canis GB1 in this study.
