喜树细胞悬浮培养体系的建立

摘    要:以喜树的幼嫩叶片为材料,探讨了MS、SH、B5基本培养基和不同激素(2,4-D、NAA、6-BA)配比对愈伤组织诱导及继代培养的影响,并筛选喜树细胞悬浮培养体系的最优培养基配方.结果表明,喜树幼嫩叶片外植体诱导愈伤培养以SH+2.0mg/LNAA+0.5mg/L6-BA为最优,愈伤组织诱导率为92%.继代培养以MS+2.0mg/L6-BA+0.5mg/LNAA为最适,在此条件下诱导的愈伤组织质地疏松,适于进行悬浮培养.

Abstract:The young leaves and stems of Camptotheca acuminata were used as the materials to study the effects of basic media and the phytohormone combination on callus induction and subculture of the callus and screen out the best cultural formula for cell suspension culture system of Camptotheca acuminata. The results showed that most suitable medium for callus induction was SH with 2.0 mg/L NAA and 0.5 mg/L 6-BA, and the callus inducing rate was 92%. MS medium with 2.0 mg/L 6-BA and 0.5 mg/L NAA was found to be the best subculture medium formula, and the callus induced under these conditions and it was suitable for cell suspension culture.
