花蕾型金银花同源四倍体的诱导和鉴定

摘    要:目的]为选育金银花优良品种提供材料。方法]以当年生银翠蕾嫩枝茎段为外植体,进行愈伤组织和不定芽培养,对获得的不定芽采用秋水仙素浸泡法和培养基中添加秋水仙素法诱导多倍体,并进行染色体鉴定。结果]染色体观察结果表明,7:30 a.m.取材,用0.2%的秋水仙素溶液浸泡3 h,用1.0 mol/L盐酸溶液在60℃下处理18 min的材料中,中期细胞多,染色体分散和着色均较好,而且可以看到清晰的点状染色体。染色体鉴定结果表明,秋水仙李浓度对多倍体诱导率有显著影响,用0.4%秋水仙李浸泡16 h的诱导率为50.0%,在秋水仙李浓度为1.0 g/L的培养基中培养8 d的诱导率为43.8%。结论]该研究为金银花同源四倍体的诱导与鉴定提供了一定的试验依据。

Abstract:Objective] The aim was to provide material for breeding good varieties of honeysuckle.Method] With the stem segments of one-year Lonicera macranthoides "silver-green" tender branches as explants,the tissue cultures of callus and adventitious bud were conducted.The polyploidies were induced from the obtained adventitious buds by the methods of soaking in colchicine solution and adding colchicine into medium and their chromosomes were identified.Result] The observation on the chromosome showed that in the material sampled at 7:30 a.m.,soaked in 0.2% colchicine solution for 3 h and treated with 1.0 mol/L chloride acid solution at 60 ℃ for 18 min,there were many metaphase cells,in which the chromosome separated and was colored better and the clear dot chromosome could be seen.The results from chromosome identification showed that the colchicine concn.had significant effects on the induction rate of polyploidy.The induction rate of soaking in 0.4% colchicine solution for 16 h was 50.0% and that of culture in medium with 1.0 g/L colchicine for 8 d was 43.8%.Conclusion] The research provided some experimental basis for the induction and identification of honeysuckle autotetraploid.
