松乳菇固态发酵棉粕脱毒条件的研究

摘    要:目的]对松乳菇固态发酵棉粕脱毒的发酵条件进行优化,以期获得最佳的发酵工艺参数。方法]采用单因素单因子法研究了碳源、氮源、无机盐含量以及温度、起始pH、料液比、接种量等因素对游离棉酚降解率的影响,在此基础上,采用响应面法评价了发酵温度、起始pH、料液比、接种量及其交互作用对游离棉酚降解率的影响,用Design-Expert 8.0.5b软件分析试验数据,建立了松乳菇固体发酵棉粕发酵条件的数学模型。结果]研究表明,适宜的发酵培养基组成为:棉粕与麦麸的比例6∶4,(NH4)2SO40.75%,KH2PO40.15%,MgSO4·7H2O 0.05%;适宜的发酵条件为:温度28℃,初始pH 6.6,料液比1∶1.4 g/ml,接种量10%。优化后的发酵条件为:温度28℃,初始pH 6.6,固水比1∶1.38 g/ml,接种量10.38%。在此条件下,游离棉酚的降解率达到87.702 3%。结论]研究可为棉籽饼粕的深度开发和生产棉籽蛋白提供参考依据。

Studies on Detoxification Conditions of Cottonseed Cake by Solid-state Fermentation with Lactarius deliciosus

Abstract:Objective] To optimize conditions of Lactarius deliciosus solid-fermentation for cottonseed meal detoxification,so as to obtain the optimum fermentation technique parameters.Method] The effects of fermentation conditions such as carbon sources,nitrogen sources,the content of nitrogen source,the content of inorganic salt,cultural temperature,initial pH value,the ratio of material to water,inoculum size on the rate of free gossypol degradation were studied by single-factor experiment.Then,the influence of temperature,initial pH value,the ratio of material to water,inoculum size and their interactions on the degradation rate of free gossypol was evaluated by using response surface methodology.Data were analyzed with Design-Expert 8.0.5b software.Result] The results showed that the optimal fermentation medium component were as follows:the ratio of cottonseed meal and wheat bran was 6∶4,(NH4)2 SO4 0.75%,KH2 PO4 0.15%,MgSO4 · 7H2O 0.05%.The optimal fermentation conditions were:temperature 28 ℃,initial pH value 6.5,the ratio of material to water was 1 ∶ 1.4 g/ml,inoculum size 10%.The optimized fermentation conditions were:temperature 28 ℃,initial pH value 6.6,the ratio of material to water was 1∶1.38g/ml,inoculum size 10.38%.Under these conditions,the rate of free gossypol degradation reached 87.702 3%.Conclusion] The study will provide a reference basis for deeply development of cottonseed meal and production of cottonseed protein.
