电针对糖尿病胃轻瘫大鼠胃窦组织IGF-1及EGF表达的影响

摘    要:目的 通过观察电针“梁门”“足三里”“三阴交”穴对糖尿病胃轻瘫(diabetic gastroparesis,DGP)大鼠胃窦组织胰岛素样生长因子1(insulin-like growth factor 1,IGF-1)及表皮生长因子(epidermal growth factor,EGF)表达的影响,探讨电针治疗DGP的可能机制。方法 选取10只正常SD大鼠为空白组,其余大鼠造模成功后选取30只随机分为模型组、电针组、胃复安组,每组10只。采用单次腹腔注射链脲佐菌素,并连续8周不规则喂养高糖高脂饲料方法制备DGP模型,每周测量血糖、尿糖值,记录症状积分。电针组大鼠取穴“梁门”“足三里”“三阴交”,胃复安组予1.7%胃复安药液灌胃(1 mL/100 g)。治疗结束后,ELISA法检测IGF-1及EGF表达。结果 与空白组相比,模型组大鼠血糖、尿糖值和症状积分显著升高(P<0.01),胃窦部IGF-1的含量明显升高(P<0.05),EGF的含量明显降低(P<0.05);经治疗后,与模型组大鼠相比,电针组和胃复安组大鼠尿糖值、症状积分、胃窦部IGF-1含量明显降低(P<0.05或P<0.01),EGF含量明显升高(P<0.05或P<0.01),电针组大鼠血糖明显降低(P<0.05),胃复安组血糖无明显变化(P>0.05),电针组与胃复安组大鼠比较,各指标均无明显差异(P>0.05)。结论 电针“梁门”“足三里”“三阴交”穴能够降低DGP大鼠血糖、尿糖值,改善DGP大鼠症状,其机制可能与电针调控胃窦组织中IGF-1与EGF的含量有关。

Effects of Electroacupuncture on Expression of IGF-1 and EGF in Gastric Antrum of Diabetic Gastroparesis Rats

Abstract:Objective To observe effects of electroacupuncture (EA) at "Liangmen (ST 21)", "Zusanli (ST 36)" and "Sanyingjiao (SP 6)" on expression of insulin-like growth factor 1 (IGF-1) and epidermal growth factor (EGF) in diabetic gastroparesis (DGP) rats, so as to explore the mechanism of EA in the treatment of DGP. Methods Ten Sprague-Dawley (SD) rats were taken as blank group. The other rats were randomly divided into model group, EA group, and metoclopramide group after successful modeling, with 10 rats in each group. DGP models were established by single intraperitoneal injection of streptozotocin (STZ) and feeding the rats with high fat-sugar forage irregularly for 8 weeks. Symptom score was recorded, and blood glucose and urine glucose were measured every week. "Liangmen (ST 21)", "Zusanli (ST 36)" and "Sanyingjiao (SP 6)" were selected in the EA group. The metoclopramide group was given 1.7% metoclopramide (1 mL/100 g) by gavage. After the treatment, the expression of IGF-1 and EGF was detected by ELISA. Results Compared with the blank group, the blood glucose, urine glucose and symptom score in the model group were significantly increased (P<0.01), the content of IGF-1 in gastric antrum was significantly increased (P<0.05), while the content of EGF was significantly decreased (P<0.05). Compared with the model group, the urine glucose and symptom score and the content of IGF-1 in gastric antrum were obviously decreased after the treatment in the EA group and the metoclopramide group (P<0.05 or P<0.01), while the content of EGF was obviously increased (P<0.05 or P<0.01). The blood glucose of rats in the EA group was significantly decreased (P<0.05), but there was no significant change of that in the metoclopramide group (P>0.05). And there was no significant difference in each index between the EA group and the metoclopramide group (P>0.05). Conclusion EA at "Liangmen (ST 21)", "Zusanli (ST 36)" and "Sanyingjiao (SP 6)" can decrease blood glucose and urine glucose, and improve symptoms in DGP rats, which may be closely associated with its effect of regulating the contents of IGF-1 and EGF in gastric antrum tissue.
