传染性法氏囊病病毒VP2基因在家蚕中的表达

摘    要:传染性法氏囊病病毒(IBDV)能引起以淋巴细胞衰竭为特征的抑制性疾病。IBDV的主要结构蛋白VP2蛋白是主要的宿主保护性抗原,在VP2上的点突变是造成IBDV抗原漂变的主要原因。为研究VP2作为IBDV亚单位疫苗的潜力,本实验将IBDV JD1株的VP2基因重组于家蚕杆状病毒转移载体pBacPAK8中,将重组载体pBacPAK-VP2与病毒Bm-Bac-PAK6的线性化DNA共转染家蚕培养细胞,获得重组病毒BacPAK-VP2。DIG点杂交表明重组病毒基因组中含有VP2基因。重组病毒感染家蚕5龄幼虫后,用ELISA、SDS-PAGE和Western blotting检测蚕血淋巴中重组VP2蛋白的相对表达量及免疫反应性。结果表明重组VP2蛋白具有免疫反应性,感染后5-6d在蚕血淋巴中的表达量最高。

Abstract:The major structure protein of infectious bursal disease virus, VP2, is the major protective of the virus and the point mutations in the variant area located in VP2 lead to the antigen shift and the sepuenced circumvent of vaccination. In this study a VP2 gene was inserted into a Bombyx mori baculovirus transfer vector pBacPAK8 and cotransfected with linear virus Bm BacPAK6 into BmN cells. The BmN cells and the silkworm of fifth instar were infected by the recombinant virus. DNA extracted from the infected cells and the immunogenesis of the infected larvae's blood were examined with dot hybridization and ELISA?Western blotting, respectively. The recombinant VP2 in blood achieved the peak at 5 day post inucolation and the expression amount of the protein in the larvae was obviously higher than that in BmN cells.
