Abstract:Summary Two highly variable DNA markers were developed for Cronartium flaccidum and Peridermium pini. The markers were used to study the genetic composition of aecia of single cankers of both species. All five cankers of P. pini were homogenic and contained only one allele in both loci studied, indicating that the aecia were homozygous. The homogeneity of the cankers suggested that either genetic exchange via spermatization does not occur in autoecious P. pini populations, or that the pairing is homothallic. Two C. flaccidum cankers contained heterozygous loci. However, one canker contained genetically variable aecia indicating heterothallism, whereas the aecia of the other canker were homogenic.
