重庆山地黄牛及其杂交牛CAST基因部分片段PCR-SSCP分析

摘    要:采用PCR-SSCP技术分析了重庆山地黄牛及其杂交牛CAST基因的多态性。在所做的2对引物中研究表明:在第18外显子处没有发现多态性;在第9外显子处发现多态性,在重庆山地黄牛及其杂交牛群体中都有AA、AB、BB3种基因型。在西杂牛(重庆山地黄牛X西门塔尔牛)、红杂牛(重庆山地黄牛X红安格斯)中A为优势基因,在重庆山地黄牛中B为优势基因。通过对各遗传多态性指标分析发现,重庆山地黄牛、西杂牛、红杂牛处于平衡状态,几个品种牛踟均在0.250~0.500,处于中度多态;几个品种牛的杂合度都偏高。

Abstract:The polymorphism of CAST gene in Chongqing mountain cattle and its hybrids was analyzed by PCR-SSCP.In two pairs of primers,there were no polymorphism detected by the 18th exon of the bovine CAST and there were three genotypes(AA,AB and BB) detected by the 9th exon of the bovine CAST in all cattle.The frequency of A allele was higher than that of B allele in Xiza cattle(Chongqing cattle × Simmental)and Hongza(Chongqing cattle ×Angus) cattle breeds,in Chongqing cattle,the frequency of B allele was higher than A allele.All three breeds were at Hardy-Weinberg equilibrium.In Chongqing cattle,Xiza cattle,Hongza cattle,the polymorphisms information contents were moderate polymorphisms(0.250
