摘    要:目的]表达和纯化拟南芥热激因子HSF1。方法]以构建的能表达热激因子HSF1的大肠杆菌Escherichia coli M15(pQE32/His6-HSF1,pREP4)为材料,用异丙基硫代-β—D-半乳糖苷(IPTG)诱导表达HSF1,再通过镍亲和层析纯化表达的HSF1,通过变性的聚丙酰胺(SDS.PAGE)电泳分析表达蛋白和纯化蛋白。结果]试验获得了表达的HSF1,并且进一步获得了纯化的HSF1。结论]该研究为探讨拟南芥HSF1在基因组的结合位点提供了试验材料,为全面认识HSF1作用机理和生理功能奠定了基础。

Abstract:Objective] This study was to express and purify Arabidopsis thaliana heat shock factor HSF1.Method] Using Escherichia coli M15 harboring HSF1(pQE32/His6-HSF1,pREP4) as experimental materials,HSF1 was induced to express with isopropyl-β-D-galactoside(IPTG);then the expression product was purified using Ni-NTA-agarose affinity chromatography and analyzed by SDS-PAGE.Result] HSF1 of Arabidopsis thaliana was successfully expressed and purified.Conclusion] This study provided materials for understanding the binding site of HSF1 on Arabidopsis thaliana chromosome,further laying a good foundation for revealing the regulatory mechanism and physiological function of HSF1.
