秋水仙碱应用于小豆诱导变异初探

摘    要:将秋水仙碱浓度为0.2000%、0.1000%、0.0500%、0.0250%、0.0125%的水溶液分别处理萌动的京农5号小豆种子8、16、24 h,通过对M1世代植株成活率、株高降低比例、M2世代变幅及变异系数、诱导变异遗传力、M2世代出现的明显变异类型进行分析,探讨秋水仙碱诱变小豆的适宜浓度及处理时间。结果表明:(1)根据M1世代相对成株率,较适宜的诱变处理为0.2000%浓度8 h和0.1000%浓度24 h;(2)处理浓度对M1世代出苗和成株率的直接影响明显大于处理时间;(3)以50%相对成株率为标准时,8、16、24 h浸泡,所适宜的处理浓度分别为0.215%、0.156%、0.115%;(4)如以株高降低50%~60%为标准,则适宜浓度为0.0500%~0.2000%;(5)M2世代中,出现花粒和感白粉病等多种变异类型。

Abstract:Sprouting seeds of Jingnong No.5 adzuki bean were treated with colchicine solution at 0.2000%,0.1000%,0.0500%,0.0250%,0.0125% and soaked for 8 h,16 h,24 h,respectively.The suitable time and concentration for inducing variations of adzuki bean with colchicine were probed into by the analysis of alive plant rate and plant height reduction rate in M_1 progeny,and by the analysis of range and variation coefficient and habitability of induced variations and obvious type of variation in M_(2)progeny.The results showed that:(1)On the basis of the alive plant rate,8 h 0.2000% and 24 h 0.1000% were the comparatively suitable treatment to induced variations;(2)Directeffect of concentration of treatment was obvious bigger than time of treatment;(3)As alive plant rate of 50% being the standard,the suitable concentrations were 0.2150%,0.156% and 0.115% in soaked times of 8 h,16 h and 24 h,respectively;(4)As plant height reducing for 50%~60% being the standard,the suitable concentrations were 0.0500%~0.2000% in the soaked times respectively;(5)There were two kinds of obvious variations of infected with powdery mildew and red-green piebald seed in M_(2)progeny,and its treatments were 8 h 0.0500% and 24 h 0.1000%,respectively.
