摘    要:油菜品种的分子鉴别在油菜分子育种和种质资源管理中有重要意义。利用RAPD分子标记技术对5个双低油菜品种或杂种(湘油15号、湘油11号、湘农油571、杂695、杂814)进行品种鉴定,建立了适合油菜的RAPD反应体系:30μL反应体系中含50 ng模板,40 pmol/L引物,150μmol/L dNTP,1.5 UTaq DNA Polymerase,2.5 mmol/L Mg2+,35.5℃的退火温度。找到了可快速鉴别5个油菜品种或杂种的两条随机引物(S151和S369),为这几个油菜品种的分子鉴别找到依据。

Abstract:Molecular identification of rapeseed varieties has important significance in the molecular breeding and germplasm resource management of rapeseed. Five double low rapeseed varieties, i. e. Xiangyou No. 15, XiangYou No. 11, Xiangnongyou 571, Za695 and Za814, have been identified by RAPD technique. The optimal RAPD reaction conditions for rapeseed were 50 ng DNA and 40 pmol/L primer, 150 μmol/L dNTP, 1.5 UTaq DNA polymerase, 2.5 mmol/L Mg^2+ in 30 μL reaction system. Two random primers, S151 and S369, could used to effectively and quickly identify the five rapeseed cultivars, which provided the basis for molecular identification of the tested rapeseed varieties.
