苦瓜多糖的测定方法研究

摘    要:为开发和利用苦瓜多糖,选用苯酚硫酸法和蒽酮硫酸法测定苦瓜多糖的含量;比较不同成熟度苦瓜中多糖的含量差别;比较冻融法制备的苦瓜汁粉及苦瓜渣粉中多糖含量。结果显示,对同一种样品利用苯酚硫酸法和蒽酮硫酸法测定,测定结果分别为28.00%和29.25%,苯酚硫酸法测定结果更为准确合理,可作为苦瓜中多糖含量测定的首选方法;测定不同成熟度苦瓜的多糖含量,发现苦瓜成熟度越高,多糖含量越低,说明苦瓜生理成熟阶段,苦瓜多糖是重要的呼吸基质之一;利用冻融法制备的苦瓜汁粉多糖含量为31.25%,苦瓜渣粉多糖含量为28.64%,说明冻融法获得的苦瓜汁没有起到完全分离苦瓜多糖的作用。

Abstract:In order to develop and use of polysaccharide of Momordica charantia,the content of polysaccharides in Momordica charantia is determined through phenol-vitriolic colorimetry method and anthrone-sulphuric acid colorimetry method;contents of polysaccharides in Momordica charantia in different maturity are compared;contents of polysaccharides in Momordica charantia in juice and dross are compared through freezing and thawing method.It indicates that,for the same sample,the results are respectively 28.00%and 29.25%through phenol-vitriolic colorimetry method and anthrone-sulphuric acid colorimetry method.The result of phenol-vitriolic colorimetry method is more reliable and accurate,it can be considered as the optimum assay method for polysaccharides in Momordica charantia.The result from the different maturity experiment shows that the higher degree of maturity,the lower the content of polysaccharide and it also demonstrates that polysaccharide is one of important substance of respiration during the phase of physical maturity.The contents of polysaccharide in juice and dross of Momordica charantia are 31.25%and 28.64%by freezing and thawing method and they illustrate that the polysaccharide in juice is separated incompletely by freezing and thawing method.
