大豆不同基因型胚尖不定芽的诱导及对抗生素的敏感性

摘    要:以大豆胚尖为外植体,在MS+3.0mg/L6BA上培养诱导8个基因型产生不定芽,筛选适合于胚尖再生系统的大豆基因型。同时,测定吉林40在诱导胚尖不定芽时对卡那霉素和头孢霉素的耐受性。结果表明,在8个大豆基因型中,2个基因型的不定芽诱导率大于90%,综合考虑不定芽诱导率和芽数两个性状,吉林40和黑农37适合于大豆胚尖不定芽再生系统。在培养基中加入卡那霉素或头孢霉素时,随着抗生素浓度的增加,胚尖不定芽诱导率急剧下降。经方差分析,卡那霉素和头孢霉素对大豆胚尖不定芽诱导率和芽数均有明显的抑制作用。在培养基内加入200mg/L卡那霉素或300mg/L头孢霉素时,胚尖不定芽诱导率和芽数显著地低于对照。在胚尖再生系统遗传转化中,建议以200mg/L卡那霉素作为抗性筛选浓度,使用头孢霉素作为脱菌剂时,浓度低于300mg/L不会降低大豆胚尖不定芽的诱导率和芽数。

Abstract:A total of eight soybean genotypes were induced to produce adventitious shoots from embryonic tips as explants in the culture media of MS +3.0mg/L 6BA in order to screen the soybean genotypes optimal for embryonic tip regeneration system,and the endurance of the induced shoots of Jiyu40 to kanamycin and cefotaximin were also studied. The results showed that the inductivity of two genotypes were higher than 90% among the eight genotypes,Jiyu 40 and Heinong 37 were fit for soybean embryonic tip regeneration system according to inductivity of adventi-tious shoots combined with the number of shoots. The inductivity of adventitious shoots decreased sharply with the increase of concentration of kanamycin or cefotaximin added in the culture media. According to the variance analy-sis,kanamycin and cefotaximin had apparent inhibition on inductivity and number of adventitious shoots,which were low compared with control when 200 mg/L of kanamycin or 300 mg/L cefotaximin were added in the culture media.The concentration of 200 mg/L kanamycin was advised to be resistance screening in embryonic tip regeneration sys-tem. The cefotaxinun whose concentration is lower than 300 mg/L as de - bacterium agent will not reduce the induc-tivity and number of adventitious shoots.
