| 摘 要： | 平滑肌细胞是胃动力的基本单位,为研究奶牛皱胃弛缓的发病机理,本研究建立了奶牛皱胃平滑肌细胞的体外培养方法。手术取奶牛皱胃组织,镜下分离出肌层,运用酶消化法分离皱胃平滑肌细胞并进行体外培养,在倒置显微镜下观察细胞的生长状态,用免疫组化染色方法进行平滑肌细胞鉴定。结果显示细胞生长状态良好,呈现平滑肌细胞特征性的"峰-谷"状结构,免疫组化染色显示细胞浆内平滑肌肌动蛋白呈阳性表达,表明该细胞为平滑肌细胞,可用于后续的试验研究。

| Abstract: | Smooth muscle cell is the fundamental unit of gastric motility.This paper established a method for cultivating the abomasal smooth muscle cells of dairy cows in vitro for researching the pathogenesis of cow abomasul atony.The abomasums were got by surgery and the smooth muscle was segregated under the microscope.The digestion method was applied to isolate the smooth muscle cells,and then these cells were prepared for culture in vitro.The cultured cells were observed under the inverted microscope,and identified by immunohistochemistry method.The results showed that the cells grew well with the typically ＂peak and valley＂ growth pattern,and expressed positive α-SM-actin in cytoplasm by immunohistochemisty.All these indicate that the cells are smooth muscle cells and can be used for further experimental study. |
