籽用乌桕优良单株遗传分子标记及亲缘关系分析

摘    要:乌桕Sapium sebiferum(L.)Roxb.]是我国极富经济价值的优良树种。在前期安徽籽用乌桕良种选育的基础上,进一步采用ISSR分子标记方法,对优良单株的遗传分子标记及亲缘关系进行分析。结果表明,筛选的10条引物在6种供试材料中均能稳定、清晰的扩增出条带,6个样品遗传多样性丰富,初步建立了5个乌桕优良单株的遗传分子标记。进一步聚类分析表明,形态学表现性状相近的乌桕优良单株,在分子生物学上的亲缘关系也较近,合适的引物能将品质优劣的籽用乌桕品种区分开来,表明用ISSR分子标记来鉴定乌桕优良品系的方法是可行的。

Abstract:Sapium sebiferum is an excellent tree species with great economic value in China. Based on the previous studies of the superior individual selections of Anhui Province, ISSR molecular marker method was adopted in this research. Molecular markers and phylogenetic relationships of the superior individuals were analyzed. The results were as follows: all of the selected 10 primers were able to amplify stable and clear bands, of which, 6 samples were abundant in genetic diversity. The molecular markers for 5 selected superior individuals were established. Phylogenetic analysis indicated that the individuals that are similar in their morphological characteristics were also closer in the molecular phylogenetic tree. Suitable primers were able to distinguish S. sebiferum varieties. The results showed that using ISSR molecular marker method to identify superior varieties of S. sebiferum is feasible.
