| 摘 要： | 为探明小檗碱对脂多糖（LPS）诱导外周血多形核白细胞（polymorphonuclear leukocytes,PMN）跨内皮细胞迁移（transendothelial migration,TEM）的影响。通过体外培养大鼠肠黏膜微血管内皮细胞,利用Tran-swell培养系统模拟内皮屏障,观察1μg/mL LPS刺激及5μg/mL小檗碱干预时,6 h后穿越内皮屏障PMN数量的变化。结果显示,LPS致炎诱导了PMN的TEM增加,小檗碱干预则能显著抑制LPS的诱导作用。试验表明,抑制PMN的TEM是小檗碱治疗LPS性炎症的机制之一。

| Abstract: | This study aimed to the effect of berberine on the LPS-induced transendothelial migration(TEM) of peripheral blood polymorphonuclear leukocytes(PMN).Rat intestinal mucosal microvascular endothelial cells were cultured in vitro,and were grown on Transwell to establish the simulated endothelial barrier.The numbers of PMN across the endothelial barrier over 6 h when 1 μg/mL LPS stimulation and 5 μg/mL berberine intervention was were investigated.The results indicated that the TEM of PMN was increased by LPS,and that the intervention with berberine could significantly inhibited the induction of LPS.This study coucluded that the inhibition of the TEM of PMN was one mechanism of berberine in treating LPS-induced inflammation. |
