| 摘 要： | 目的]制备罗非鱼鱼皮胶原蛋白多肽，并对其理化性质进行研究。方法]以从罗非鱼鱼皮中提取的胶原蛋白对原料，分离制备活性肽，并测定其理化性质。结果]利用胰蛋白酶在45℃、pH值8、加酶量5％条件下，酶解3h，进而用葡聚糖凝胶G-50通过蛋白质纯化仪进行分离，纯化得到较纯的多肽Y32。电泳得知Y32分子量为4．0456kDa；DSC820e测定Y32的变性温度为57．5℃；调节pH值测定其等电点为8．7；用氨基酸组成分析仪测定Y32氨基酸种类及其含量发现，Y32多肽中共含有19中氨基酸，其中，甘氨酸所占比例最大，符合胶原蛋白氨基酸的基本构成。结论]为进一步开发利用罗非鱼鱼皮提供了基础资料。

| Abstract: | Objective] The research aimed to prepare the tilapia skin collagen and study the physicochemical characteridtics.Method] Taking the tilapia skin collagen as raw materials,the isolation of bioactive peptides and its physicochemical characteristics were studied.Result] The collagen was hydrolyzed by trypsin.The temperature,pH value and the amount of enzyme were 45 ℃,8 and 5% respectively.The purer peptides Y32 was obtained by purification with Sephadex G-50.The molecular weight of Y32 was 4.045 6 kDa thought SDS-PAGE.The denatured temperature measured by DSC820e was 57.5 ℃,and the pH value of Y32 were 8.7.The determination results of the amino acid species and the contents of Y32 determined by amino acid composition analyzer showed that there were 19 kinds of amino acids in Y32.Thereinto,the ratio of glycin was the biggest.And Y32 was in accord with the basic compostion of amino acid.Conclusion] The study provided the foundation data for developing and utilizing the tilapia skin deeply. |
