康乃馨脱除斑驳病毒组织培养技术研究

摘    要:作者对康乃馨进行脱除斑驳病毒培育无毒苗的技术研究。试验表明:用0.3mm茎尖接种,在MS+BA0.7mg/L+NAA0.1~0.2mg/L培养基上愈伤组织诱导率为90.0%;在MS+BA0.8mg/L+NAA0.2mg/L培养基上愈伤组织分化出芽率为74.0%,平均每块愈伤组织有芽5.9个。在MS+BA2.0mg/L+NAA0.3mg/L和MS+BA2.0mg/L+NAA0.5mg/L培养基上增殖倍数6.5~7.4;在1/2MS+NAA0.01mg/L+IBA0.05mg/L上诱导生根率为90.0%;降低培养温度、增加光照强度及培养基中加入20万单位青霉素可减轻玻璃苗现象。经汁液传染试验检测,组培苗脱毒率为65.0%。

Abstract:the result showed that a length of 0.3 mm stem tips was inoculated.Callus induction rate is 90.0% in culture medium of MS+BA0.7mg/L+NAA0.1-0.2mg/L,Callus germination rate is 74.0%in MS+ BA0.8mg/L+NAA0.2 mg/L,and there are 5.9 buds per callus,propagation multiple is 6.5~7.4 of culture medium in MS+BA2.0mg/L+NAA0.5mg/L,induction rooting rate is 90%in1/2MS+NAA0.01 mg/L+ IBA0.05 mg/L,Reduce incubation temperature,increasing light intensity and medium supplemented with 200 000 units of penicillin can reduce vitrification phenomenon.Tissue culture seedling removal rate is 65.0% by the sap transmission test.
