| 摘 要： | 目的]探讨一种提取盐芥高质量DNA的方法。方法]分别采用简化SDS法、改良CTAB法、CTAB法和改良尿素法提取盐芥DNA,测定不同方法所提取DNA的纯度和浓度,并进行电泳和酶切检测,并对其结果进行比较。结果]改良CTAB法提取的DNA纯度高、质量好,没有糖类、酚类及蛋白质的污染,能用于PCR扩增和限制性内切酶酶切。CTAB法提取的DNA没有蛋白质的污染,但有酚类物质的污染,可作为PCR扩增的模板。SDS法以及尿素法提取的DNA质量较差,不能满足分子生物学研究的要求。结论]改良CTAB法比其他方法更适合分子生物学分析和研究。

| Abstract: | Objective] The aim of the research was to discuss a kind of method for extracting high-quality DNA from Thellungiella salsuginea.Method] DNA was extracted from T.salsuginea by simplified SDS method,improved CATB method,CTAB method and improved urea method respectively.The purity and concentration of DNA extracted by different methods were determined.And they were detected by electrophoresis and restriction enzyme digestion and their results were compared.Result] DNA extracted by improved CTAB method had high purity and good quality,without the pollution of carbohydrate,phenols and protein.It could be used in PCR amplification and digestion with restriction enzymes and the improved CTAB method was more accordant with the demands of molecular biology research than other methods.DNA extracted by CTAB method was not polluted by protein,but it was polluted by phenols.It could be taken as the templates for PCR amplification.The quality of DNA extracted by SDS method and urea method was so worse that it could not meet with the demands of molecular biology research.Conclusion] Compared with other methods,the improved CATB method was more suitable for molecular biology research. |
