摘    要:目的]筛选适宜原球茎一步成苗的培养基,探讨原球茎试管苗的快繁移栽技术。方法]以大花蕙兰(Cymbidium grandiflorum)幼嫩芽为外植体进行组织培养研究。以MS为基本培养基附加不同的激素,分别设计5种配方的原球茎诱导培养基和3种配方的原球茎一步成苗培养基,原球茎一步成苗的最适培养基。用农用链霉素稀释2500~3500倍对原球茎试管进行消毒后,栽入不同的基质,研究原球茎试管苗的移栽技术。结果]大花惠兰在MS+6!BA1.0mg/L+NAA0.1mg/L配方培养基上诱导原球茎效果最好。试管苗的快繁以MS+6-BA2.0mg/L+NAA0.2mg/L的培养基为最适,21d后原球茎大量增殖和分化,诱导出丛生芽并生根。在腐殖土中适当使用农用链霉素有利于练苗成功。结论]该试验探究出原球茎从增殖,诱导分化到生根只用一种培养基,大大缩短生产周期,但其生根率只有75%,需做进一步研究。

Abstract:Objective The aim of the research was to screen medium suitable for one-step-seedling formation and discuss transplanting technique.Method In the tissue culture Cymbidium grandiflorum,with the tender bud of C.grandiflorum as explant,Ms as the basic medium was added with the different hormones,5 formulas of protocorm induction media and 3 formulas of protocorm one-step-seedling formation media were designed to screen the optimum medium suitable for one-step-seedling formation .The tub seedlings of protocorm were sterilized with agricultural streptomycin diluted at 2 300~3 500 times,then transplanted on the different matrixes to study the transplanting technique of protocorm tub seedlings.Result The formula medium of MS+ 6-BA 1.0mg/L + NAA 0.1mg/L got best induction effect on for protocorm induction of C.grandiflorum.The formula medium of MS+6-BA 2.0 mg/L + NAA 0.2 mg/L was most suitable to the rip reproduction of protocorm tub seedlings.After 3 weeks of culture,the protocorm could proliferated and differentiated to induce the clustered shoots and roots.The tub seedlings of protocorm were sterilized with agricultural streptomycin and transplanted on the humus soil could obtain success of seedling training Conclusion Using only one kind medium from proliferation,induction and differentiation was studied in this test,which shortened the production period.But the rooting rate was only 75 % got in this medium that should need to study further.
