水稻OsBTB蛋白在非亲和/亲和互作中表达方式的初步研究

摘    要:为了解水稻OsBTB蛋白在非亲和/亲和互作中的表达情况,构建含OsBTB全长基因的原核表达载体,经诱导表达及纯化获得OsBTB蛋白。以OsBTB蛋白为抗原制备兔多克隆抗体,经间接ELISA法测定抗体效价为1∶2000。Westernblot分析确认该抗体与OsBTB蛋白可以特异结合。应用该抗体检测受不同稻瘟病菌(Magnaporthegrisea)生理小种侵染的相应水稻(Oryzasativa)品系中OsBTB蛋白的表达,结果表明OsBTB蛋白的表达方式呈多样性,该蛋白在非亲和反应中的表达时间明显早于其在亲和反应中的表达。免疫胶体金技术确认OsBTB蛋白存在于细胞核内。

Abstract:To understand the expression of rice OsBTB protein in the incompatible/compatible interactions between rice and Magnaporthe grisea, recombinant prokaryotic expression vector containing full-length sequence of open reading frame from OsBTB gene was constructed. Polyclonal anti-OsBTB rabbit antibody has been successfully prepared by using purified OsBTB protein as immunogen. The titer of antiserum against OsBTB was about 1:2000 using ELISA. Western blotting analysis showed that the antiserum could be binded to the expression OsBTB protein specifically. The OsBTB expression pattern in different rice (Oryza sativa) lines induced by corresponding Magnaporthe grisea isolates displayed diversities when detected by using anti-OsBTB antiserum. OsBTB protein was expressed earlier noticeably in incompatible interaction than that of compatible. Immuno-gold labeling test was performed, and it was found that the protein localized in the nuclei of M. grisea infected rice cells
