摘    要:为探讨鹿蹄草、功劳叶和地榆三种中草药对大肠杆菌的抑制活性及作用机理。采用菌落计数法测定中草药提取液对大肠杆菌的抑制作用;采用倍比稀释法测定中草药的最小抑菌浓度(MIC)和最小杀菌浓度(MBC);通过紫外分光光度计测定不同时间段菌液吸光值和电导率值;将大肠杆菌在带药营养肉汤培养基中培养10 h并在扫描电子显微镜(SEM)下观察大肠杆菌的超微结构变化。结果表明:三种中草药均具有较好的抑菌作用,且中草药浓度越高,抑制作用越强。鹿蹄草的MIC值为31.25 mg/mL,MBC值为62.50 mg/mL;功劳叶和地榆的MIC值均为62.50 mg/mL,MBC值均为125 mg/mL。加药组的大肠杆菌生长受到抑制,菌体细胞膜渗透性增加。扫描电镜显示大肠杆菌细胞出现变长现象,表面出现许多凹坑。三种中草药均有较强的抑菌作用,其中鹿蹄草的抑菌效果最明显。

Inhibitory Effect and Antimicrobial Mechanism of Three Kinds of Chinese Herbal Medicine on Escherichia Coli
Abstract:The present work aimed to explore the antibacterial effect and potential inhibitory mechanism of three kind of Chinese herbal medicine against Escherichia coli. The experiment used plate count method to test the extract of herbal medicine on the inhibitory effect of Escherichia coli, and utilized doubling dilution method to determine the minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). At different time periods, the absorbance values of bacteria liquid and conductivity of Escherichia coli were determined by Ultraviolet spectrophotometer. After 10 hours cultured in a nutrient broth with drugs, through scanning electron microscopy (SEM) to understand the effect of three kinds of Chinese herbal medicine on ultrastructural of Escherichia coli. The result showed that all the herbal medicine displayed a better antibacterial effect against Escherichia coli. The MIC value of Pyrola was 31.25 mg/mL and its MBC value was 62.50 mg/mL; the MIC value of others were both 62.50 mg/mL and the MBC value were 125 mg/mL. It can be discovered that the growth of Escherichia coli in dosing group was restrained and its permeability of cytomembrane was increased. The observations by SEM revealed that Escherichia coli cells got longer and cell surface appeared a lot of pits. It can be concluded that three kinds of Chinese herbal medicine had strong antibacterial effect and Pyrola was more effective against Escherichia coli.
