江南牡丹叶片耐热差异蛋白的鉴定与分析

摘    要:为探究江南牡丹在高温胁迫下的应答模式,以进一步获得可能的耐热目的蛋白,本研究以江南牡丹耐热品种‘羽红’的小苗为实验材料,运用同位素标记相对定量与绝对定量技术(iTRAQ),结合质谱鉴定与相应生物学分析,探究40℃高温处理和25℃正常生长的江南牡丹叶片中蛋白差异表达情况。一共鉴定得到的蛋白总数1 916个,大于等于1.5倍的差异蛋白有100个,表达上调的有55个,表达下调的有45个。基因本体(GO)分子功能分析发现差异蛋白富集前三的分别是分子功能中的催化活性和结合功能以及生物学进程中的代谢过程。KEGG通路分析发现差异蛋白富集程度最高的是包括丙酮酸代谢、碳代谢等过程的代谢通路,其次是内质网中的蛋白质合成和光合作用通路。同时发现了肽基脯氨酰基异构酶、APX、HSPs、FBA、CaM、RubisCo、乙酰辅酶A羧化酶、GS等蛋白可能是江南牡丹应对高温胁迫中发挥重要作用的目的蛋白。本研究结果为进一步探究江南牡丹在高温胁迫下的应答模式提供了理论基础。

Abstract:In order to explore the response pattern of tree peony under high temperature stress,and further obtain the possible heat-resistant target protein,the’Yuhong’was used as experimental materials in this study.Isobaric tags for relative and absolute quantification(i TRAQ)were combined with mass spectrometry identification and corresponding biological analysis to investigate normal growth at 25℃and high temperature treatment at 40℃.Total of 1916 proteins were found,and 100 differentially expressed proteins were more than or equal to 1.5-fold,including 55 up-regulated and 45 down-regulated.GO molecular function analysis showed that the first three proteins were enriched by catalytic activity and binding function in molecular function and metabolic process in biological process.KEGG pathway analysis found that the highest levels of differential protein enrichment were among the metabolic pathways including pyruvate metabolism,carbon metabolism and other processes,followed by protein synthesis and photosynthesis pathways in the endoplasmic reticulum.The study found that proteins such as peptidylprolyl isomerase,APX,HSPs,FBA,CaM,RubisCo,acetyl-CoA carboxylase,GS and so on may be the target proteins of tree peony that play an important role in high temperature stress.The results in this study will provide a basis for further exploring the molecular regulation mechanism of tree peony under high temperature stress.
