摘    要:由猪胸膜肺炎放线杆菌(APP)引起的猪传染性胸膜肺炎是猪最重要的呼吸道传染病之一。为研究该细菌表面蛋白的结构与功能,选取APP代表菌株,提取其基因组DNA,用TSPS091随机消化成3~8kb的片段并回收,连接到预先消化的入ZAPⅡ载体上,经过包装、扩增和滴定后,成功地构建了APP基因组文库。

Abstract:Porcine contagious pleuropneumoniae, which was one of the most important infectious respiratory diseases, was caused by actinobacillus pleuropneumoniae. In order to study the structures and functions of APP surface proteins, the genome library was constructed. The genome was extracted from the representative APP strain, and randomly digested by TSP5091 into 3~8 kb fragments. The genome library was successfully constructed by ligation of recovered fragments to predigested lambda ZAPII after packaging, amplification and titration.
