天麻不同种质过氧化物同工酶分析

摘    要:采用不连续聚丙烯酰胺凝胶垂直平板电泳技术,对陕西汉王略阳中药科技有限公司天麻基地的3个天麻不同种质,同一生长期花茎中过氧化物酶(POD)同工酶酶谱进行比较研究及相似性系数数据处理分析.结果显示:不同天麻种质过氧化物同工酶活性不同,且各具有特征谱带,其中卵果天麻、绿秆天麻、红秆天麻的酶谱条带数分别为7条、8条和9条.从POD酶谱上分析,卵果天麻最原始,红秆天麻分化程度较大,是3种种质中最为进化的一种.研究表明过氧化物酶同工酶谱在天麻不同种质之间,有着很高的多态性,除少数共有的特征谱带外,各个种质之间存在较明显的谱带差异.

Abstract:The aim of the research was to discuss the genetic relationship of different Gastrodia elata germplasms cultivated in Lueyang county. The POD isozyme system in the scapes of three Gastrodia elata germplasm cultivated in Lueyang county was studied by means of vertical polyacrylamide gel e-lectrophoresis. The results indicated that the three germplasm presented different peroxidase isozyme activities, and had their own key bands. Gastrodia elata Bl. f. elata, Gastrodia elata Bl. f. viridis Makino, Gastrodia elata Bl. var. obovata sp nov. respectively had nine, eight, and seven isozyme bands. The results also showed that Gastrodia elata Bl. var. obovata sp nov. was the earliest germplasm and Gastrodia elata Bl. f. elata was the most evolving germplasm in the three. POD zymogram belts of different germplasm had abundant polymorphism , which indicated that different populations of Gastrodia elata was distinguishing on the genetic essence. And the difference of the POD isozyme zymogram among different germplasms was obvious except for a few common characteristic zymogram belts.
