栉孔扇贝和虾夷扇贝通用微卫星引物的筛选及其在杂种鉴定中的应用

摘    要:目的]为进行大规模栉孔扇贝×虾夷扇贝群体的杂种鉴定以及家系中的亲子鉴定提供了基础。方法]将已发表的25对虾夷扇贝的微卫星引物在栉孔扇贝基因组DNA上进行PCR扩增,并利用筛选出的通用引物对栉孔扇贝(♀)×虾夷扇贝(♂)进行杂种鉴定。结果]通过PCR扩增,筛选出6对引物获得扩增产物,其中2对表现为多态性。将这6对具有属间通用性的微卫星引物对栉孔扇贝(♀)×虾夷扇贝(♂)进行PCR扩增,均能扩增出特异性条带,其中引物P13F449和KMY134的扩增产物中可以明显地分辨出来自父母本群体的特有条带,从而确定了杂交子代的杂种身份。结论]虾夷扇贝和栉孔扇贝间通用微卫星引物是存在的,而且部分引物可以直接用于属间杂种的鉴定。

Abstract:Objective] The study aimed to provide the base for hybrid identification of Patinopecten yessoensis×Chlamys farreri population and paternity testing in family in great scale.Method] Twenty-five pairs of reported microsatellite primers from P.yessoensis were made for PCR amplification on the genome DNA of C.farreri and the hybrid identification of C.farreri(♀)×P.yessoensis(♂) were made by using the selected universal microsatellite primers.Result] Through PCR amplification,six pairs of primers were selected and their amplifying products were obtained,with 2 pairs being polymorphic.When C.farreri(♀)× P.yessoensis(♂) were tested by using 6 pairs of microsatellite primers which could be applied to both the two kinds of scallops,the specific bands could be amplified from each one of the 6 pairs,among which,the specific bands from both agnate and maternal colony could be distinguished obviously in the amplified products of P13F449 and KMY134,and accordingly the hybrid identity of hybrid generations could be affirmed.Conclusion] It was proved that there was certain microsatellite primers that could be applied to both C.farreri and P. yessoensis and some of the primers could be directly used for the identification of intergeneric hybrid.
