杜仲籽粑的深度开发利用研究

摘    要:目的]研究从杜仲翅果中提取亚麻酸之后的籽粑中制备桃叶珊瑚苷及其苷元的工艺方法。方法]采用硅胶柱层析法从杜仲粕中分离纯化桃叶珊瑚苷,优化柱层析分离、纯化条件,利用酶解技术选择最佳酶解条件,运用高效液相色谱法(HPLC)对产品进行检测分析。结果]最佳上样量为2%,使用V(甲醇)∶V(石油醚)∶V(乙酸乙酯)为4∶0.5∶9的混合溶剂进行洗脱,通过收集、重结晶可获纯度为97%的桃叶珊瑚苷产品,利用酶解技术在30 min时桃叶珊瑚苷能达到最佳的酶解效果。结论]该研究为高效制备桃叶珊瑚苷及其苷元的工业生产提供依据。

Study on the Deeply Efficient Utility of the Crude Extract of Eucommia ulmoides Seeds

Abstract:Objective] To obtain the optimized purification of aucubin (Au) and aucubigenin from the crude extract of the residue of Eucommia ulmoides seed meal left after linolenic acid extraction.Method] The optimal conditions of column chromatography for aucubin and of enzymolysis for aucubigenin were investigated,and the products were detected by HPLC.Result] An optimal purification of Au was achieved by using a 2% mass of silica gel for aucubin adsorption followed by desorption with a methanol/petroleum ether/ethyl acetate mixture (4∶0.5∶9,V/V).The eluate fractions were harvested and recrystalized,and a final aucubin product with 97% purity was obtained.The best time of enzymolysis was 30 min.Conclusion] This study will provide the reference for commercial process of aucubin and aucubigenin.
