胭脂鱼精子低温保存及其在人工授精中的应用

摘    要:目的]研究胭脂鱼精子的低温保存方法,为胭脂鱼的人工繁殖和人工授精提供基础依据。方法]试验配制了A、B、C、D4种精子保存液,以0.7%的NaCl溶液作为对照,以精子被激活后的活率为指标,对4种精子保存液保存成熟的胭脂鱼精子的效果进行评价。结果]在低温(0~4℃)条件下,4种精子保存液之间及其与对照溶液之间的差异比较明显。其中保存效果最好的是B液,能在120h内使精子的活率保持为80%;D液次之,能在84h内使精子的活率保持为80%。胭脂鱼在淡水中快速运动时间为(24.0±3.5)s,寿命为(128.0±9.5)s。2007~2008年精子保存B液在胭脂鱼的人工授精中进行应用,受精率为53%~85%。结论]用精子保存液低温保存胭脂鱼精子的方法是切实可行的。

Study on the Application of Spermatozoa Preservation at Low Temperature in Artificial Fecundation of Myxocyprinus asiaticus Spermatozoa

Abstract:Objective] The study aimed to investigate the method of low temperature preservation for Myxocyprinus asiaticus spermatozoa and provide the foundation data for artificial reproduction and fecundation.Method] In the test,4 kinds of spermatozoa preservative fluids of A,B,C and D were prepared,with 0.7% NaCl solution as CK.The effects of four preservative fluids of mature spermatozoa from M.asiaticus were evaluated with the fast moving rate of spermatozoa after being motivated as an index.Result] Under the low temperature of 0-4 ℃,the differences between 4 kinds of the preservative fluids and between them and CK fluids were more obvious.Among them,B fluid was the best,which could keep 80% of spermatozoa having fast moving ability within 120 h and D fluid was second,which could keep 80% of spermatozoa having fast moving ability within 84 h.The moving time of M.asiaticus spermatozoa in fresh water was(24.0±3.5) s,with the life time of(128.0±9.5) s.From 2007 to 2008,when the permatozoa preservative B fluid was used in the artificial insemination of M.asiaticus,the fecundation was 53%-85%.Conclusion] The method of keeping M.asiaticus spermatozoa at low temperature with spermatozoa preservative fluids was feasible.
