高效液相色谱法检测鳗鱼中孔雀石绿及其代谢物残留

摘    要:目的]采用高效液相色谱法测定鳗鱼中孔雀石绿及其代谢物隐色孔雀石绿残留量。方法]鳗鱼样品用乙腈提取,经脱脂、浓缩、净化和流动相溶解,用高效液相色谱二极管阵列检测器(DAD)测定。结果]在HPLC条件下,14min内可完成对孔雀石绿和隐色孔雀石绿的测定。在0~0.1μg/ml浓度范围内,孔雀石绿和隐色孔雀石标样工作曲线的相关系数(r)分别为0.9999和0.9998;在2、25、100μg/kg添加水平条件下,孔雀石绿、隐色孔雀石绿的加标回收率分别为85.7%~89.7%和87.6%~91.2%,测定相对标准偏差(RSD)小于5%(n=5);该方法对孔雀石绿和隐色孔雀石绿的检测限分别为1和2μg/kg(S/N≥3)。结论]该研究应用HPLC测定鳗鱼中孔雀石绿等残留准确、可靠,其灵敏度符合法规要求。
