白灵菇液体培养工艺研究

摘    要:目的]为白灵菇进一步开发提供科学依据。方法]采用单因素试验和正交试验,确定白灵菇液体培养的最适碳源、氮源和最佳培养条件。结果]适合白灵菇液体培养的碳源是玉米粉和葡萄糖,氮源是黄豆粉和蛋白胨。碳源和氮源的主次因素为黄豆粉>葡萄糖>玉米粉>蛋白胨。最佳碳、氮源组合为玉米粉3.0%,葡萄糖1.5%,黄豆粉2.0%,蛋白胨0.2%,生物量为1.781g/100ml。在培养条件各因素中,主次因素为pH值>接种量>转速>装液量。pH值对菌丝生物量有极其显著的影响,接种量、转速、装液量的影响不明显。结论]最佳培养条件是摇瓶装液量80ml/250ml,接种量10%,pH值6.5,摇瓶转速180r/min,25℃培养8d,生物量为1.792g/100ml。

Abstract:Objective The research aimed to provide scientific basis for further development of Pleurotus nebrodensis.Method The optimum carbon source,nitrogen source and the best culture conditions were confirmed through single factor test and orthogonal test.Result The suitable carbon source for liquid culture of P.nebrodensis was corn meal and glucose,the suitable nitrogen source was bean flour and peptone.The factors of carbon source and nitrogen were in sequence as:bean flour > glucose > corn meal > peptone.The optimum combination of carbon source and nitrogen source was 3 % corn meal,1.5 % glucose,2 % bean flour and 0.2 % peptone,with the biomass of 1.781 g/100 ml.The culture conditions were in sequence as:pH value> inoculation amount > rotational speed > liquid filling amount.pH value had an extremely significant effects on the mycelial biomass,while inoculation amount,rotational speed and liquid filling amount had not obvious effect.Conclusion The optimum culture conditions were as follows:Liquid filling amount per shake flask of 80 ml/250 ml,inoculation amount of 10 %,pH value of 6.5,rotational speed of shake flake of 180 r/min,culturing 8 days at 25 ℃,with biomass of 1.792 g/100 ml.
