摘    要:旨在找到一种简便的SNP检测方法用于水稻种性鉴定及遗传多样性分析,本研究以245份长江中下游主推杂交水稻品种及亲本为研究材料,从1319个SNP位点中筛选出124对多态性高的位点,建立SNP的高效检测体系。选用其中的一个标记sf0141821553和SSR的标记RM7120来检测水稻的纯度,分别利用Caps-AccI标记和SNP-sf0601764762扩增分析亲本及杂交后代Wx的基因型,两者结果完全一致,说明水稻功能性SNP标记完全可用于水稻种品纯度和种真实性的鉴定。用124对位点对245份杂交水稻进行遗传多样性分析,聚类分析显示,245个品种间的遗传相似系数在0.64~0.97之间,以遗传相似系数0.64为阈值,可以将245个水稻品种分为两个类群,这两个类群分别在遗传相似系数为0.712和0.667处又可以分为两个亚群,结果表明材料间存在明显的群体结构,能精确区分待测品种的基因型以及品种间的遗传差异。利用聚丙烯酰胺凝胶电泳来实现SNP标记的检测,方法简便且不需要大型仪器设备和昂贵的试剂,便于实验室开展水稻品种的鉴定工作。

A Method for Detecting SNP Markers Based on PCR in Rice
Abstract:This study aims to find a simple SNPs detection method for rice species identification and genetic diversity analysis. 245 main hybrid rice varieties and their parents widely cultivated in the middle- lower reaches of the Yangtze River were used as materials. 124 pairs of highly polymorphic loci were screened out from 1319 SNP loci and an efficient SNP detection system was constructed. One of the selected SNP markers, sf0141821553, and a SSR maker RM7120 were used to identify the purity of hybrid seed. The genotypes of Wx in parental species and hybrid generation groups were amplified by Caps-AccI and SNP-sf0601764762, respectively, and they had the same result. The genetic diversity analysis was conducted with the 124 pairs of loci among the 245 rice varieties. The clustering analysis revealed that the genetic similarity coefficients within the 245 varieties were from 0.64 to 0.97. The 245 varieties were classified into two cluster groups with the similarity coefficient of 0.64. Each of the two cluster groups was further divided into two subgroups with the similarity coefficient of 0.712 and 0.667 respectively. These results showed obvious population structure among these rice varieties, and the genotypes of the tested varieties and the genetic differences among varieties were accurately distinguished. The detection of SNP markers by polyacrylamide gel electrophoresis is simple, and does not require large instruments and expensive reagents, which is convenient for laboratory identification of rice varieties.
