超低温冷冻对俄罗斯鲟精子抗氧化酶活性的影响

摘    要:为了探讨超低温(-196℃)对精子的损伤机理,采用试剂盒测定了冷冻前后俄罗斯鲟精浆和精子中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH—PX)和谷胱甘肽还原酶(GR)等酶活性。结果表明,经过超低温冷冻保存后,俄罗斯鲟精子活力下降,添加抗冻保护液冷冻后的精浆中SOD、CAT、GSH—PX活性较鲜精组显著升高(P〈0.05),较未添加保护液组显著降低(P〈0.05),GR的活性则显著低于鲜精组(P〈0.05),但显著高于未添加保护液组(P〈0.05);添加抗冻保护液冷冻后的精子中SOD、CAT、GSH—PX活性较鲜精组显著降低(P〈0.05),较未添加保护液组显著升高(P〈0.05),GR活性则显著高于鲜精组(P〈0.05),但显著低于未添加保护液组(P〈0.05)。这说明超低温冷冻对俄罗斯鲟精子的抗氧化系统产生了较大影响,成为影响精子活力的一个重要原因。

Abstract:The effects of cryopreservation(-196 ℃)on the antioxidant activities of enzymes (SOD, CAT, GSH-PX and GR) in seminal plasma and spermatozoa of Russian sturgeon Acipenser gueldenstaedti were studied by a reagent kit to clarify damage mechanism of spermatozoa. The spermatozoa were found to show lower motility after preserved compared to the fresh semen, and the activities of SOD,CAT,GSH-PX in seminal plasma supplemented with cryoprotectants increased significantly (P<0.05)compared to the control group (P<0.05). The activity of GR showed significantly lower compared to the fresh seminal plasma(P<0.05),but significantly higher compared to the seminal plasma without supplementation of the cryoprotectants (P<0.05).Compared to the fresh sperm, the spermatozoa supplementation of the cryoprotectants had lower activities of SOD,CAT,GSH-PX(P<0.05).However, the significantly higher activity of GR was observed in the spermatozoa supplementation of the cryoprotectants compared to the fresh sperm(P<0.05),and the significantly lower activity of GR was found in the seminal plasma without supplementation of the cryoprotectants (P<0.05). The results showed that cryopreservation had significant effects on anti-oxidative system in the spermatozoa of Russian sturgeon and the motility of the spermatozoa.
