基于mtDNA Cyt b基因全序列探讨岩扇贝与3种扇贝的遗传变异水平及其亲缘关系

摘    要:为探讨新引进种岩扇贝Crassadoma gigantea与国内3种主要经济扇贝(虾夷扇贝Patinopecten yessoensis、栉孔扇贝Chlamys farreri和海湾扇贝Argopecten irradians)的遗传变异水平及其之间的亲缘关系,通过PCR扩增及测序获得4种扇贝的mtDNA Cyt b基因全序列,并对其序列组成、遗传多样性、遗传距离和系统发育等进行了分析。结果表明:4种扇贝的单倍型数、多态性位点数、单倍型多样性指数、核苷酸多样性指数和平均核苷酸差异数,分别在3~4、2~15、0.545~0.800、0.001 00~0.005 72和0.788~6.545之间,表现出较低的遗传变异水平;4种扇贝的种间遗传距离为0.255~0.429,其中,岩扇贝与虾夷扇贝的遗传距离最近(0.255),与海湾扇贝的遗传距离最远(0.429);系统进化树分析显示,岩扇贝与虾夷扇贝独自聚为一支,表明二者之间的亲缘关系较近。本研究结果将为岩扇贝的引种与养殖以及今后的遗传育种研究工作提供基础信息。

Genetic variability and relationship between rock scallop Crassadoma gigantea and three other scallops based on sequence of mtDNA Cyt b gene

Abstract:The sequences of Cyt b gene were amplified by PCR and sequenced to obtain the whole sequences of Cyt b gene in 10 individuals sampled randomly from four scallop species including rock scallop Crassadoma gigantea, bay scallop Argopecten irradians,Zhikong scallop Chlamys farreri and yesso scallop Patinopecten yessoensis to under-stand the genetic variation and the relationships among the four scallop species. Then, the related analysis of se-quence characterization, genetic diversity, genetic distance and phylogenetic evolution were analyzed by related software. The results showed that,the sequences of Cyt b gene in Zhikong scallop, rock scallop, bay scallop and yesso scallop were shown more than 99% homologous to the corresponding sequences in GenBank, with haplotype number of 3-4,polymorphic sites of 2-15,haplotype diversity of 0.545-0.800,nucleotide diversity of 0.001 00-0.005 72 and average nucleotide difference of 0.788-6.545,indicating that the maximal genetic diversity(0.429) between rock scallop and bay scallop and the minimal(0.255) between rock scallop and yesso scallop.The phyloge-ny trees further confirmed that there was close genetic relationship between rock scallop and yesso scallop.The find-ing provides basic information for future research on species introduction and intensive culture,germplasm resources management,and genetics and breeding in rock scallop.
