摘    要:对目前生产上主栽籼型杂交水稻亲本的组织培养特性进行了系统研究,发现优质杂交籼稻丰优香占的恢复系父本R6547的组培特性与粳稻模式品种相似.愈伤组织诱导频率达95%以上,其愈伤组织诱导、继代培养和分化再生水平远高于绝大多数籼稻品种。该品种愈伤组织分化再生频率高达87%;继代4次仍有95%的愈伤组织保持胚性状态,表现出很强的耐继代能力;在用农杆菌介导法转移外源基因时,相对地保持了其优良的组培特性。进一步讨论了利用R6547建立高效转化系统的可能性及其重要意义。

Abstract:The characteristic of in vitro tissue culture in parents of the major cultivated hybrid cultivars of indica rice was systematically studied. Results showed that R6547, the restorer parent of a famous hybrid rice cultivar FengVouXiangZhan, had excellent tissue culture characteristic similar to the japonica rice Zhonghua 11 and better than most indica cultivars in capabilities of callus induction, subculture tolerance and plantlet differentiation. The frequency of callus induction from mature and immature embryos of R6547 was above 95 % and the ratio of plantlet regeneration from its embryogenetic callus reached at 87%. Through successive four times of subculture, the 95% of its calli were still in the embryogenetic condition, reflecting strong capability of subculture tolerance. When an alien gene was transferred into R6547 with the Agrobacterium tumefaciens transformation, the excellent tissue culture characteristic of R6547 was maintained. The importance and feasibility of establishing a highly efficient in vitro tissue culture and transformation system in indica rice with R6547 were discussed.
