栎属植物DNA提取方法的研究

摘    要:为了获取高质量的栎属植物基因组DNA,采用经过改良的SDS法和CTAB法对几种常见栎属植物进行基因组DNA的提取.结果表明:改进的SDS法和CTAB法都较常规的方法好.所提出的DNA较纯净.改进的CTAB法与改进的SDS法比较.改进的CTAB效果较好,所获得的DNA经紫外分光光度计和琼脂糖凝胶电泳检测,纯度较高,OD260/OD280为1.75~1.80,运用SSR和RAPD进行分析时,可以获得较好的扩增片段,说明蛋白质、多糖、RNA等去除较干净.

Abstract:In order to obtain high quality genomic DNA,comparison of the methods for isolating genomic DNA from the genus Quercus L.plants was studied.The results showed that both modified SDS(sodium dodecy sulphate) and CTAB(cetyl triethl am-monium bromide) methods for isolating genomic DNA from the genus Quercus L.plants were better than the original SDS and CTAB methods.DNA yield of modified CTAB method was higher than that of modified SDS method.The isolated DNA was exam-ined by ultraviolet absorption test and aga...
