Abstract:As the endocrine factor, incretin play a pivotal role in the regulation of glucose and lipid metabolism. As an important incretin, GIP are involved in glucose uptake and lipid deposition. To investigate the role of GIP in the common carp (Cyprinus carpio), a series of experiment was performed. The common carp gip were cloned from the foregut by RT-PCR, and the sequence was analyzed by bioinformatics and the mRNA expression was detected by Real-time PCR. The result showed that the ORF of common carp gip were 324 bp, which encode 107 amino acids. Based on amino acids sequence, the structure of GIP precursor protein contains four domains: signal peptide, N-terminal domain, mature GIP peptide and C-terminal domain. The results of sequence alignment and phylogenetic tree showed that the common carp GIP protein were closest to that of zebrafish. The Real-time PCR results indicated that the gip was widely expressed in multiple tissues, and was with higher expression in the intestine and pituitary of common carp. In OGTT experiment, the gip mRNA expression was significantly increased in common carp foregut by glucose treatment for 1 h and 2 h. In the fasting and re-feed experiment, the gip mRNA expression in the foregut was markedly decreased after fasting 7 days, and was significantly increased after re-feed. In the feeding experiment, the gip expression was increased in the high glucose and high lipid groups. The results will provide the data base for consummating GIP biological functions and theoretical foundation for investigating the regulatory of glucose and lipid metabolism in fish.
