摘    要:采用PCR-RFLP方法,对采自安徽怀远、河南范县及安徽舒城的3个群体共112尾泥鳅(Misgurnus anguilli-caudatus)线粒体DNA(mtDNA)的D-loop区段进行PCR扩增,使用11种核酸限制性内切酶进行酶切。结果显示,在11种内切酶中,8种有酶切位点,4种存在个体间变异,共产生7种单倍型。怀远、范县和舒城群体的核苷酸多样性指数为0.0033、0.0083、0.0011,单倍型多样性指数分别为0.6048、0.8370、0.2344。范县与怀远、舒城群体间的Rogers遗传距离分别为0.3456和0.6847,怀远和舒城群体间的Rogers遗传距离为0.6326。SspⅠ的酶切类型C组成的单倍型在舒城群体中占87.5%,而在范县群体中为0;同时发现,SspⅠ的酶切类型C、D和XspⅠ的酶切类型B分别组成的单倍型仅在范县群体中存在。

PCR-RFLP Analysis on mtDNA D-loop Region of Three Populations of Misgurnus anguillicaudatus
Abstract:PCR amplication and 11 restriction endonucleases were conducted on the D-loop region on the mitochondria DNA of 112 loaches(Misgurnus anguillicaudatus) from three populations in Huaiyuan of Anhui,Fan county of Henan and Shu city of Anhui.According to restriction endonucleases,among those 11 endonucleases,eight restriction enzyme cutting sites,four individual variation and seven haplotypes were observed.Nucleotide diversity index(h) and haplotype diversity index(π) in Huaiyuan,Fan county and Shu city populations were 0.0033,0.0083,0.0011 and 0.6048,0.8370,0.2344,respectively.The Rogers' genetic distances were 0.3456,0.6847,0.6326 between Fanxian population and Huaiyuan population,Fan coutny population and Shu city population,Huaiyuan population and Shu city population respectively.The haplotype composed of SspⅠ's C patternccounting for 87.5% in Shu city population,but it was not detected in Fanxian population.Meanwhile,the haplotypes composed of SspⅠ's C and D pattern and XspⅠ's B pattern only existed in Fan county population.
